Sepragen Corporation has been helping customers with their custom chromatography needs for over a decade. No two chromatography processes are alike. Some require low flow rates, some high flow rates; some use alcohol or other flammable solvents and require explosion proof design and certification; others need to run in a cold room; some need stand alone control while others may need to communicate with an in-house SCADA or DCS; and some may require a polymeric or even a disposable flow-path while others may require a robust stainless steel construction. In such cases when of the shelf solutions don’t work, customers often go down the trap of having either their engineers design their own systems in house or having their engineering companies outsource the skid fabrication from third party fabricators. Experience has shown that climbing out of this slippery slope is difficult at best. While the initial objectives of making clinical lots are easily met with a less than state of the art design, it is in the reuse of the systems down the road that the problem occurs. Frequently if the process changes are made that make the skids unusable the “original hardware engineer or “software engineer” or A&E company are gone and there is no competent authority to implement the changes. Often the documentation is not clear and change control is not easy. No one is at hand to do rev control on the software of a specialized machine and keep up with the version changes in the operating system. Often no one is available for support, calibration and trouble- shooting or training. Eventually skids like this are “mothballed” and long term value is sacrificed at the altar of short term convenience or control.

Sepragen through its experience has developed a tool- box of technologies that provides you, our customer, several unique benefits in terms of product capability and time tested reliable, rapid execution. Some of these hardware sub-systems and there advantages are stated below:
  1. High precision, positive displacement pumping systems with 100:1 turn down ratio that enable a wide range of operation.
  2. NEMA 4X purged enclosures and pneumatically
  3. Electronic auto air ejection system that eliminates accidental air from getting into the column
  4. Low hold up design sanitary valve manifolds for buffer selection, fraction collection and column switching
  5. Multi-mode detector flow cell where all sensors like UV or pH are located thereby eliminating sensor lag.
  6. Leak sensor that can sense leaks in plumbing assembly or pumps
Sepragen Quantasep Software

Sepragen’s state of the art, intuitive software platform has been designed with configurability and customization in mind. The software has been designed to accept hardware changes such as the addition or elimination of valves, pumps, sensors etc and still have interface-ability to all devices. The software contains the ability to do both manual and automated runs and view chromatograms and their analysysis and get batch reports. Data can easily be backed up to a Windows server. Also, the Sepragen software can be configured to communicate with plant level systems like Delta V via OPC server. Menus can be designed and changed easily to accommodate process changes. Generation upgrades are routinely done and version control and traceability maintained over continuing use. The software can be turned off and the control handled to a central controller if needed.

Examples of Custom designed Chromatography Skids

System specifications have usually come from the customers or A&E companies. These have usually been refined after discussions with our team to minimize cost and time of delivery and ensure greater reliability and support. Some common examples of custom skids are listed below:
  1. Gradient Skid NEMA 4X, Class 1 Div II construction with 4 inlets on each pump, 6 outlets rated for 40 LPM with pre and post column pH and conductivity sensors, pre- column filter, dual wavelength UV in 316 SS construction with TUV certification.
  2. Gradient System with 316 SS Flow path with ATEX classification of Class I Div I T4 BASEEFA rating, pressure rating of 50 psi with 6 inlets on each of the two pumps along with 10 fractions, bubble trap, temperature, conductivity, pH, pressure, air, leak and multi- wavelength UV sensing.
  3. Isocratic system with 60 LPM with 4 inlets and 2 outlets. Communication with customer network.
  4. All polymeric flow-path, 30 LPM dual pump system with inlets, outlets, column forward, bypass and reverse capability and fraction collection. Unique ability to control column pressure and inlet pressure head to pumps. Cold room compatible operation.
  5. System with built in buffer blending capability.
  6. System with removable polymeric flow- path.
